CHI是菊科多年生草本植物,其嫩叶及利用其肉质根软化栽培获得的芽球均可食用,是新兴的特种蔬菜.CHI耐涝性差,生长期若田间积水则易引起霉烂.目前,对CHI的研究主要集中在栽培方面,对其离体培养方面的报道较少.本试验从激素等几个方面对CHI进行离体再生体系的研究,其结果如下:
(1)CHI四种外植体以A和B做外植体的处理效果好于以C和D做外植体的处理.
(2)CHI脱分化容易.单加E或F均能较快速的启动CHI外植体的脱分化.但单加E其愈伤再分化效果好于单加F的效果.单加F诱导的愈伤易分化出大量的根.而在单加的E中,又以F诱导的菊苣外植体再分化效果较好.研究表明,当F为3.0mg/L、G为0.3 mg/L时对诱导CHI愈伤发生及分化的效果最好.
(3)在用不同品种对CHI愈伤诱导分化效应的研究中发现,H品种愈伤的诱导分化效果优于I品种.
(4)本试验采用的3种基本培养基对CHI进行不定芽的直接诱导.随着培养基中无机元素种类和浓度的减少,对CHI不定芽的诱导率也随之减少:M培养基诱导效果最好,S培养基其次,在W培养基上没有诱导出不定芽.
(2)在不添加任何激素的M培养基的基础上,CHI不同部位诱导不定芽的能力差异较大:J和K能诱导出不定芽,而L不能诱导出不定芽,并且,R的诱导率高于P的诱导率.
3 CHI组培苗生根较容易,但浓度过高会抑制其生根.Q诱导生根的效果优于R,最佳生根培养基为:M
